Canine Distemper: RT-PCR Versus the Rapid Test
Short answer
For suspected canine distemper, the rapid test gives a quick first look, RT-PCR looks for the virus's genetic material and can find small amounts, and the ELISA adds information from the immune side. Vets often read them together.

For a dog with suspected distemper, the rapid test gives a quick first look, RT-PCR looks for the virus's genetic material and can find small amounts, and the ELISA adds information from the immune side. No single test settles every case, so a vet reads them together with the dog's signs and vaccine history.
What signs may point towards distemper?
Canine distemper is caused by a virus that mostly affects puppies and dogs that are not fully protected. It spreads through coughs and through discharge from the eyes and nose. Common signs include watery or sticky discharge from the eyes and nose, a cough, fever, a poor appetite and loose motions. Some dogs later show nerve signs, such as twitching or fits.
These signs overlap with kennel cough, parvo and other illnesses, so the signs alone cannot name the cause. A sick puppy should see a vet promptly. Please do not wait for a lab result before getting help.
What does the rapid test do?
The Canine Distemper Virus (Rapid Test) is a small kit that looks for pieces of the virus, called antigens. It is usually done on a swab or discharge sample, and the lab confirms the sample needs when you book.
Its strength is speed: it gives the vet a first look. Its limit is that it needs enough virus in the sample. A negative kit may miss a dog that carries only a little virus, or one sampled early or late in the illness.
What does RT-PCR add?
The Canine Distemper RT-PCR is the RT-PCR (real-time PCR) option. It searches for the distemper virus by its genetic material. Because the test makes many copies of a small stretch of that material before reading it, it can find the virus when only a little is present. The lab confirms which sample is needed when you book.
Very sensitive tests can sometimes pick up vaccine virus in a dog that was vaccinated recently, so tell your vet about any recent vaccination.
When a dog's signs could fit more than one virus, the Dog Viral RT-PCR Panel may help. It typically groups RT-PCR tests for several dog viruses, and the exact list is confirmed when you book.
What does the ELISA add?
The Canine Distemper ELISA Test is done on a plate in a laboratory. Depending on the kit, it looks for parts of the virus or for antibodies, the proteins a dog's body makes when it meets a germ. The lab confirms what the kit looks for when you book.
If it is an antibody test, remember that antibodies also appear after vaccination and after past exposure. A result of that kind alone cannot show that a dog is ill now. Your vet uses it to understand the immune side of the picture, not as a stand-alone verdict.
Why might a vet use more than one test?
Each test is strong in a different way, and weak in another.
- The rapid kit is quick, but it can miss a low amount of virus.
- RT-PCR is more sensitive, but it has more laboratory steps and, like any test, depends on a good sample.
- The ELISA shows something about the immune response, but an antibody result is not proof of current illness.
Timing also matters. Eye and nose signs often come first, and nerve signs can show up later, sometimes when the virus is hard to find with a swab. At that stage the vet may lean on the examination and other tests instead of repeating the same one. A registered veterinarian puts all of this together with the dog's age, vaccine history and signs.
Distemper spreads between dogs, so ask your vet how to keep a sick dog away from others while the cause is checked.
